Annex 1 Compliance Checklist for Australian Sterile Manufacturers
TAKEWAY
This checklist highlights the areas most likely to create gaps during an Annex 1, CCS or TGA readiness review.

Use this checklist before your next Annex 1 readiness review
Australian sterile manufacturers are under increasing pressure to show that contamination risks are understood, controlled and actively managed.
Contamination Control Strategy
Confirm that your CCS connects facility design, process controls, cleaning, disinfection, environmental monitoring, utilities, personnel flows, deviations and CAPA into one coherent control framework. The CCS should be specific enough to support inspection questions, change control and periodic review.
Facility and equipment design
Review whether cleanroom layouts, pressure cascades, material transfers, RABS, isolators, single-use assemblies and fill-finish equipment reduce contamination risk by design rather than relying mainly on procedural controls.
Aseptic process controls
Check that aseptic manipulations are minimized, justified and supported by validated interventions, operator qualification, media fills, transfer controls and clear batch record evidence.
Environmental monitoring and process verification
Verify that monitoring locations, alert/action limits, trend reviews, viable and non-viable data, and investigation triggers are risk-based and linked to actual process contamination hazards.
CAPA roadmap and project delivery
Prioritize gaps by patient risk, regulatory exposure, implementation complexity and project dependency. Convert the gap list into a practical roadmap with owners, timelines, interim controls and verification steps.
